  Dr. Xu is a Senior Research Fellow in the Program inCardiovascular and Metabolic Disorders, Duke-NUS Medical School Singapore. Shereceived her Ph.D from Case Western Reserve University in 2011. From 2011-2012,she underwent postdoctoral fellowship training at Harvard Medical School inBoston, MA. Because of family reason, she moved to Singapore and continued herpostdoc training in Duke-Nus Medical School in 2013. Her research focuses onepigenetic regulation of adipocyte development.

讲座简介

   Recent years have seen an upsurge ofinterest in understanding the mechanisms underlying brown adipose tissue (BAT)development and activation. How these processes are regulated at thepost-transcriptional level, however, has yet to be fully understood. Toinvestigate roles for RNA binding proteins (RBPs) in BAT, we conducted RNA-seqto systemically profile >400 RBPs in different adipose depots, during whiteadipose tissue browning, and during brown adipogenesis, leading to theidentification of Y-box binding protein 2 (Ybx2), a BAT-enriched RBP. Knockdownof Ybx2 blocks brown adipogenesis in vitro, while its overexpressionpromotes BAT marker expression in brown and white adipocytes.  Ybx2 knockout mice can form BAT but fail toactivate a full thermogenic program. Integrative analysis of RNA-seq and RNA-immunoprecipitationfollowed by RNA-seq (RIP-Seq) revealed a set of Ybx2’s mRNA targets, includingPgc1α, that were destabilized by Ybx2 depletion. Thus, Ybx2 is a novelregulator orchestrating BAT activation.
